I load the image in from Lightroom Cloud via the “Edit In” Feature, it sends a TIFF file to Gigapixel, and never loads the image. It also loads two instances of Gigapixel, but that’s not a big deal. More importantly, the issue I’m having is that the image, despite appearing in the gigapixel library, cannot be accessed or edited in any way.

I don’t work for Topaz but, here’s the thing Adobe Lightroom or as you referred to it as Lightroom Cloud doesn’t actually use Gigapixel as a plugin not in the traditional sense it’s somewhat convoluted.
Meaning, when you send an image to Gigapixel via the Edit in Method Lightroom will create a temporary TIFF File and open Gigapixel as the Standalone version and the problem you’re experiencing is a communication issue between Lightroom and Gigapixel.
In other words, Lightroom is asking Gigapixel to do something and Gigapixel can’t hear it and seeing two instances of Gigapixel is another example of that.
Basically, the communication between the two has always been a bit fickle unreliable.
To get Gigapixel to actually work, you have to stop using the “Edit In” button and use the Manual Export Method. It’s one extra click, but it works every time.

Firstly, you need to Export your image from Lightroom by pressing the Shift plus E (Export) and choose TIFF, 16-bit, Adobe RGB. Save it to your Desktop or Folder.

Open Gigapixel separately and Drag that TIFF into Gigapixel yourself. The editing controls will now be fully active because Lightroom is no longer watching or locking at that specific file.

Save the image once you’re added your Enhancements and Upscaled the image then, drag it back into the Lightroom window to import it.
As mentioned before I realise it’s an extra step but, it’s certainly more reliable and works every time

One more thing to add: files from cloud services are not supported.
Ensure that files are stored locally on the internal or an external drive before attempting to import into Gigapixel.
