is it 4k or exceeds the size? I ask because h264 codec supports max 4k and if you have just one pixel more than 4k you must choose other codec, h265 or ProRes in TVAI.
Logs show a subtitle codec error stemming from the app trying to convert the input file.
Try swapping the Audio Mode to convert and run the export again. Might need to remux the original file and drop the subtitle tracks from the mkv if you are not keeping them with the export.
The original is a Voyager episode in SD. Trying to upscale it to 4K. I did try h264 and h265 with the same error result. Granted the original file was deinterlaced in Handbrake and saved as mp4. Will try without subtitles and see if that makes a difference.
However, I do not see an option to add or remove subtitles inside Topaz.

Correct. The limit is exactly 4096x4096 pixels for h264_nvenc encoding, and 8192x8192p for hevc_nvenc encoding, at least for Nvidia rtx 20-series and newer.
Can try sending the file out as mov or mp4 and that will drop the subtitles.
